| I. Background | |
| AHR 反應(yīng)元件(AhRE),全稱為芳香烴反應(yīng)元件,是位于特定基因上游啟動(dòng)子區(qū)的一段保守 DNA 調(diào)控序列,也是芳香烴受體信號(hào)通路的終點(diǎn)靶標(biāo)。作為細(xì)胞應(yīng)對(duì)外源性化 學(xué)信號(hào)的關(guān)鍵分子開關(guān),AhRE 構(gòu)成了生物體化學(xué)防御系統(tǒng)的核心調(diào)控節(jié)點(diǎn),在環(huán)境應(yīng)答 和毒理代謝中發(fā)揮著至關(guān)重要的作用。 AhRE 介導(dǎo)的信號(hào)傳導(dǎo)遵循經(jīng)典的配體激活模式。當(dāng)環(huán)境污染物如二噁英、多環(huán)芳烴等外源性配體進(jìn)入細(xì)胞后,會(huì)與細(xì)胞質(zhì)內(nèi)的芳香烴受體結(jié)合,激活的受體隨即轉(zhuǎn)位進(jìn)入細(xì)胞核,與芳香烴受體核轉(zhuǎn)位因子形成異二聚體。該復(fù)合物能夠特異性識(shí)別并結(jié)合 DNA 上 的 AhRE 核心序列,進(jìn)而招募共激活因子,啟動(dòng)下游基因的轉(zhuǎn)錄。被誘導(dǎo)表達(dá)的基因主要包括Ⅰ相代謝酶如 CYP1A1、CYP1A2,以及Ⅱ相代謝酶如谷胱甘肽-S-轉(zhuǎn)移酶等,這些酶 協(xié)同作用,負(fù)責(zé)對(duì)進(jìn)入體內(nèi)的外源性有毒物質(zhì)進(jìn)行代謝轉(zhuǎn)化和清除排泄。 |
