I. Background | |
B 細(xì)胞成熟抗原 (BCMA) 是一種跨膜糖蛋白,屬于腫瘤壞死因子受體(TNFR)超家族,表達(dá)于正常、惡性漿細(xì)胞和成熟 B 細(xì)胞表面,其他組織中表達(dá)量極少。BCMA 與另外兩個(gè)功能相關(guān)的TNFR 超家族成員—B 細(xì)胞活化因子受體(BAFF-R)和 TACI 一起,協(xié)同調(diào)節(jié) B 細(xì)胞增殖、成熟和存活,以及分化為漿細(xì)胞 (PC)。BCMA具有一種可溶形式 sBCMA,其通過 γ-分泌酶活性直接從膜 BCMA 脫落。sBCMA 保留了胞外結(jié)構(gòu)域和部分跨 膜區(qū)。sBCMA 是 B 細(xì)胞參與人類自身免疫性疾?。ㄈ缦到y(tǒng)性紅斑狼瘡、類風(fēng)濕性關(guān)節(jié)炎 和多發(fā)性硬化癥)的潛在生物標(biāo)志物。 BCMA 有兩種激動(dòng)劑配體:增殖誘導(dǎo)配體 (APRIL) 和 BAFF;APRIL對(duì) BCMA 的結(jié)合親和力比 BAFF高得多,并且它還能與 TACI 結(jié)合,而 BAFF對(duì)BAFF-R的選擇性更高。 配體與 BCMA結(jié)合后,多發(fā)性骨髓瘤細(xì)胞中多個(gè)生長和存活信號(hào)級(jí)聯(lián)被激活,最常見的是 活化 B 細(xì)胞的核因子 κ 輕鏈增強(qiáng)子 (NF-κB),但也包括 RAS/MAPK 和 PI3K-Akt 信號(hào)通路。這些通路通過調(diào)節(jié)細(xì)胞周期檢查點(diǎn)來刺激增殖,通過上調(diào)抗凋亡蛋白(如 Mcl-1、 BCL-2、BCL-XL)來提高存活率,以及產(chǎn)生細(xì)胞粘附分子(如 ICAM-I)、血管生成因子(如 VEGF、IL-8)和免疫抑制分子(如 IL-10、PD-L1、TGF-β)。 |
